PE-22-28
PE-22-28 is a peptide of seven amino acids made as a trimmed-down version of spadin, a 17-residue peptide that comes from the propeptide of sortilin, also known as neurotensin receptor 3. The same research group later began calling it mini-spadin.

What researchers study it for
- Pharmacology of the TREK-1 potassium channel
- Depression-related behavior tests in mice
- Neuron formation and synapse building in the hippocampus
- Stroke and depression after stroke in mice
How it is thought to work
The idea is that it blocks TREK-1, a two-pore-domain potassium channel. Patch-clamp recordings from cells carrying human TREK-1 gave an IC50 of 0.12 nM for PE-22-28, compared with 40 to 60 nM for spadin. The group later reported a two-sided action: channel activity went up at low concentrations and down at higher ones.
Where the research stands
Preclinical only. All published data come from cells and mice studied by one French laboratory. ClinicalTrials.gov lists no human trial, and the peptide is not an approved medicine.
