Semax

Semax is a seven-residue peptide (Met-Glu-His-Phe-Pro-Gly-Pro) from Russian laboratories. It joins the ACTH(4-7) stretch to a Pro-Gly-Pro ending, and papers commonly label it an ACTH(4-10) analog.

What researchers study it for

  • Rat models of reduced blood flow to the brain
  • Levels of BDNF and its receptor TrkB
  • Rodent learning and memory tasks
  • Animal models that mimic Alzheimer's disease

How it is thought to work

Rat experiments link Semax to higher BDNF and TrkB in the hippocampus and to more transcription of neurotrophin genes after induced ischemia. It has none of the hormone action of ACTH. Which receptor it acts through is unknown; in a single cell-based study it failed to switch on the melanocortin-4 receptor yet dampened the response to alpha-MSH.

Where the research stands

On Russia's drug register; not approved by FDA. Russia's official register of vital and essential medicines includes Semax. FDA has not approved it, and the clinical work behind it is largely Russian and has not been checked by outside groups.
